Leveraging deep sequencing data to move from predictive to mechanistic models of plastid gene expression

Deep sequencing approaches have drastically scaled up the identification and quantification of regu-latory events. This will enable us to build integrated models to understand the contribution of different levels of gene regulation on chloroplast gene expression, in initially two areas: Sequence-based mod-els of transcript production and changes under acclimation, and predictive models for RNA-binding pro-tein based regulation of stability and translation. Our efforts will contribute to a quantitative framework for plant gene expression and, in the long term, to the interplay of organelle and nuclear expression programs.
